关于内毒素限值的确认，求各位大神指导

关于新产品内毒素限值的确认，药典上规定的做法还是觉得有点不明白，国外药典内毒素限制怎么查，有比较给力的参考资料么？，新手，请各位大神指教哈

内毒素限值的确定药品、生物制品的细菌内毒素限值( L )一般按以下公式确定：
L = K /M
式中 L 为供试品的细菌内毒素限值，一般以EU /m l、EU/mg或E U /U (活性单位）表示；
K 为人每千克体重每小时最大可接受的内毒素剂量，以 EU/(kg * h )表示，注射剂 K = 5EU/(kg • h) ，
放射性药品注射剂K = 2. 5EU/(kg • h) ，鞘内用注射剂 K = 0. 2E U /(kg - h) ；
M 为人用每千克体重每小时的最大供试品剂量，以m l/(kg • h ) , mg/(kg . h)或 U /(k g • h )表示，人
均体重按60kg计算，人体表面积按1.62m2计算。
注射时间若不足1 小时，按1 小时计算。供试品每平方米体表面积剂量乘以0. 0 2 7即可转换为每
千克体重剂量(M ) 。
按人用剂量计算限值时，如遇特殊情况，可根据生产和临床用药实际情况做必要调整，但需说明理由。
我就想问问楼主，哪个地方没整明白？CP2015药典附录写得很清楚的。

Endotoxin Limit
The endotoxin limit for parenteral drugs, defined on the basis of dose, equals K/M♦
♦, where K is a threshold pyrogenic dose of endotoxin per kg of body weight, and M is equal to the maximum recommended bolus dose of product per kg of body
weight. When the product is to be injected at frequent intervals or infused continuously, M is the maximum total dose administered in a single hour period. The endotoxin limit for parenteral drugs is specified in the individual monograph in units such as EU/mL, EU/mg, EU/Unit of biological activity, etc.

DOSAGE AND ADMINISTRATION
If significant diarrhea occurs during therapy, this antibacterial should be discontinued.
(see WARNING box.)
INTRAMUSCULAR—Adults: Serious infections—600 mg (2 mL) intramuscularly
every 24 hours. More severe infections—600 mg (2 mL) intramuscularly every 12 hours
or more often. Pediatric patients over 1 month of age: Serious infections—one
intramuscular injection of 10 mg/kg (5 mg/lb) every 24 hours. More severe infections—
one intramuscular injection of 10 mg/kg (5 mg/lb) every 12 hours or more often.
INTRAVENOUS—Adults: The intravenous dose will be determined by the severity of
the infection. For serious infections doses of 600 mg of lincomycin (2 mL of
LINCOCIN) to 1 gram are given every 8 to 12 hours. For more severe infections these
doses may have to be increased. In life-threatening situations daily intravenous doses of
as much as 8 grams have been given. Intravenous doses are given on the basis of
1 gram of lincomycin diluted in not less than 100 mL of appropriate solution
(see PHYSICAL COMPATIBILITIES) and infused over a period of not less than
one hour.

L=k/M = (5EU/kg) / (600mg/60kg) = 0.5EU/mg
K = threshold pyrogenic dose of endotoxin per kilogram of body mass,
M = maximum recommended bolus dose of product per kilogram of body mass.

请问医疗器械的细菌内毒素限值怎么定，只知道14233.2对注射器具有要求，是20EU/件

你这计算没错，人家静脉大输液，几百ml，你才1ml，可不把标准拉低了么。

我意思是，L值跟最大供试品剂量M有关，别人如果是静脉滴注比如生理盐水，500ml，M值是你1ml的500倍，那你的L可不就比别人大500倍把标准拉低了么。
