关于小容量注射剂追加装量的疑问

记得GMP中有规定小容量注射剂在灌装过程中需要进行装量追加，对于这点有几个疑问。
1.EU和FDA是否有类似的要求？
2.此要求的根本目的主要就是为了防止传统的小容量注射剂需要抽取、配伍过程中的损耗吧？是否还有其他因素的考量？
3.现今像预灌封注射器形式的小容量注射剂，已从根本上避免了Q2中提到的那些因素，在做国内市场的产品时，是否还需参照现在的GMP进行装量的追加？

楼主指的是overfill过量灌装，还是overage过量投料？

先默认是overfill过量灌装

美国FDA指导原则《Allowable Excess Volume and Labeled Vial Fill Size in Injectable Drug and Biological Products Guidance for Industry》规定：In the case of drug products requiring reconstitution, the product should be designed to meet the label claim and acceptable overfill, and allow for correct dosing，同时，The excess volume included in a drug product should be described in the common technical document (CTD) section 3.2.P.1, Description and Composition of the Drug Product.

药典2020版通则<0102>关于注射剂过量灌装体积的规定与USP基本一致：注射剂的灌装标示量不大于50ml时，有参考原则

其目的就是为了弥补临床使用损失，楼主理解没毛病，如果可以保证临床使用需求，不进行进行过量灌装也是ok的

对于Q2 ，生产企业主要是考虑万一装少了，不合格怎么办？
可以考虑下为什么药典上对于小容量”装量“为什么规定为：依法测定（通则0102），应不低于标示量。

灌了5ml，打不出去5ml

Hi 木木老师，您引用的FDA的指导原则原文里有提到：In the case of drug products requiring reconstitution。这里的reconstitution我的理解就是像西林瓶和安瓿瓶这种不能直接一次注射的包装形式。即说明FDA也有对小容量过量灌装的类似要求。
还有如果想依靠预灌封的包装形式优点不参照相关要求进行过量灌装，在产品申报时是否需要做一些对于装量的补充研究或说明？

预充针因其极小的过量罐装，确实与普通安瓿或者西林瓶有所区别，我理解上企业应当对其最小过量罐装量进行研究，如果不进行过量罐装其临床用量也是符合要求的，可不进行过量罐装，这些研究过程需要在注册资料中展示
